My research interests are focused on combining artificial intelligence with liquid biopsies towards personalised therapeutic strategies and earlier detection and monitoring of cancer relapse. Our research has been pioneering in development and utilisation of next-generation sequencing methods for early detection and monitoring of cancer. Our research was among the first to highlight the importance of copy number aberrations AND somatic mutations in cfDNA for real-time monitoring of tumour evolution and acquired resistance to aromatase inhibitors by next-generation sequencing (A), as well as showing that cfDNA profiles can deconvolute the heterogeneity observed in single circulating tumour cells (B).

Current areas of research:

Earlier detection of acquired resistance in metastatic breast cancer using ctDNA
In collaboration with Professor Jacqui Shaw:

  • Utility of ctDNA methylation for earlier detection of impending resistance in patients with metastatic breast cancer.
  • Inferring gene expression through nucleosome occupancy in ctDNA to highlight key genes involved in acquired resistance to hormone therapies.

Utility of ctDNA for earlier detection of relapse and progression in endometrial cancer

Gynaecology research group lead by Dr Esther Moss.  I lead laboratory parts of:

  • Microsatellite loci - GutteryDetection of impending relapse or progression earlier than PET/CT scans or clinical examination through personalised NGS of ctDNA
  • Analysis of microsatellite instability in ctDNA for stratification of patients towards immunotherapies
  • Elucidating differences in mutational profiles of endometrial cancer patients based on ethnicity
  • Deconvolution of immune cell infiltration for improved therapeutic stratification using NGS.

Artificial intelligence for improved cancer diagnosis, individualised patient therapies and prediction of relapse

In collaboration with Professor Yudong Zhang:

  • Using state of the art deep neural networks for improving cancer detection in mammograms
  • Prediction of cancer molecular subtype and clinically actionable mutations using diagnostic slides
  • Prognostication of cancer patients at high-risk of relapse using diagnostic slides to improve therapeutic strategies
  • Combining state of the art deep neural networks with ctDNA for improved treatment regimens and monitoring of patients of follow-up.

SEND Study Success

Congratulations to Chris Avery, who with collaborating colleagues published findings of the SEND study, a multi-centre randomised trial using two different surgical protocols for mouth cancer treatment.  A nationwide first for this type of trial, using real-world data.  The paper, published in the British Journal of Cancer, is available by clicking this link: SEND Paper
