DNA Sequencer

The DNA sequencing service utilizes an Applied Biosystems 3730 sequencer to provide a high-throughput low-cost service available to both Internal and External clients.

The service was established in 1993 and operates on a variety of levels depending on clients needs, offering full template preparation through to analysis of sequence, or simple low-cost analysis of completed sequence reactions.

There are three different types of analysis available:


Level 1

The user supplies an overnight bacterial cell pellet or crude PCR product plus primer. PNACL purifies the plasmid or PCR product by Qiagen 9600 Robot using QIAPrep Turbo kits or QIAquick kits respectively, carries out cycle sequencing reactions, cleans them up using Centrisep columns and analyses the products using the 3730 automated sequencer.

Level 1 samples submitted to one of the sample drop-off locations prior to 14:00 will usually have data returned by 10:30 on the third working day (68.5 hour turn-around time).


Level 2

The user supplies a template and primer. PNACL carries out cycle sequencing reactions, cleans them up using DyeEx columns and analyses the products using the 3730 automated sequencer. PNACL also has a stock of common primers which can be used in for sequencing at no further cost.

Level 2 samples submitted to one of the sample drop-off locations prior to 14:00 will usually have data returned by 10:30 on the second working day (44.5 hour turn-around time).


Level 3

The user supplies completed, dried sequencing reactions from which you have removed unincorporated dye terminators. PNACL analyses the products using the 3730 automated sequencer.

Level 3 samples submitted to one of the sample drop-off locations prior to 14:00 will usually have data returned by 10:30 on the next working day (20.5 hour turn-around time).

If you choose to send your sequencing to an external company you will have to pay VAT @20%. University of Leicester users should take this into account when making a direct price comparison between PNACL and other companies.
